I think Newman's having a laugh -- "can't kiss you here, Jimbo." It all depends on what you think of Newman's humor and I think it's self-mocking and self-deprecating.
Dean makes the homosexual joke as a kind of gauntlet to Newman -- see how daring I am; can you keep up? And Newman responds with "can't here," a joke that goes along with Dean's and tries to trump it. "Can't hear" would, on the other hand, be an effort to deflect the first joke, even a nonsequitur. I think they're definitely more in tune with each other than that. I also think Newman wants to match Dean in 'daring' in making a joke about being gay because Dean already has the part and Newman, to this point the star of "The Silver Chalice" and nothing else, is trying to up his game if not in Dean's eyes than in the eyes of Elia Kazan & co.
Plus there's a Newman book coming out from the "Brando Unzipped" guy that will probably include 25 pages on the sex between Dean and Newman (and, what the hell, Kazan) that occured before and after this screen test, so the point is settled. (True story -- that the book is coming out, I mean, in July or so....)

if he meant "can't here", then the emphasis in the phrase would've slightly been on "can't"
if he meant "can't hear", then both words have the equal emphasis, which is how I heard it after 2 or 3 times.
